NovaBlue—Highly recommended for general cloning purposes. Features extremely high transformation efficiency, blue/white screening of recombinants by lacZ [alpha]-complementation, endA and recA mutations for high plasmid quality and yield, and F factor to support M13 growth and single stranded plasmid rescue of vectors carrying the f1 origin of replication.
BL21(DE3) and BL21(DE3)pLysS—Strains are widely used hosts for protein expression from pET recombinants. Suitable for production of protein from target genes cloned in pET vectors by induction with IPTG. BL21 strains are deficient in both lon and ompT proteases. Designation of DE3 indicates that the host is a lysogen of [lambda]DE3, and therefore carries a chromosomal copy of the T7 RNA polymerase gene under control of the lacUV5 promoter. Strains carrying pLysS express T7 lysozyme. This lysozome further suppresses basal expression of T7 RNA polymerase prior to induction, thus stabilizing pET recombinants encoding target proteins that affect cell growth and viability.

Cells are provided in 50µL single-use aliquots that eliminate the need to sub-aliquot, freeze or thaw, or waste partially used vials. Transformation can be done in the tube containing the cells. Two types of host strains are available: NovaBlue and BL21(DE3)/BL21(DE3)pLysS.
