BAKERBOND® is a family of silica-based products designed for optimum performance, reproducibility, and scaleability. BAKERBOND® bonded phases are synthesized through trifunctional silane chemistry to form a layer of multiple carbon-siloxane covalent bonds on the surface of the silica particles. The resulting bonded phases are polymeric type, providing increased resistance to hydrolysis, greater stability at pH extremes, and reduced silanol interactions for higher recoveries and less tailing. Trifunctional silane chemistry also provides a more consistent ligand density for increased resolution. To ensure lot-to-lot consistency, raw materials, in-process 'intermediates,' and finished products are subjected to particle size, particle size distribution, surface area, ligand loading, surface coverage, selectivity, moisture content, pH, mean pore diameter, titer (as appropriate), chromatographic use, and other tests.

All columns incorporate proprietary bonding chemistry on 5 or 15 µm wide-pore spheroidal silica gel particles with an average pore diameter of 300 Å. The protein binding capacity of these columns, as determined by static batch type extractions of standard protein solutions, is typically 0.1 to 0.2 g protein/g dry packing. These packings are chemically stable from pH 2 to 8.5. They are not damaged by the use of cleaning and antimicrobial agents such as sodium azide, dilute aqueous solutions of trifluoroacetic acid and acetic acid, organic solvents such as methanol and acetonitrile, and concentrated aqueous solutions of salts and urea.

ABx is a unique mixed-mode ion exchange medium that provides both 'affinity-like' antibody purity (70 to 99%) and fractionation in a single step. Extremely high antibody purity can be achieved by using ABx in conjunction with WP PEI (wide-pore polyethyleneimine), a weak anion exchange medium that contains primary, secondary, and tertiary amines.
