This mAb is specific to HER2 / ErbB2 and shows minimal cross-reaction with other members of the family. HER2 is a member of the EGFR family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. HER2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary.
Flow Cytometry: 0.5-1 ug/million cells in 0.1ml IF: 1-2 ug/ml IHC (FFPE): 0.5-1 ug/ml for 30 min at Room Temp (1) The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the mAb to be titered up or down for optimal performance.<p>1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.
