The ReliaPrep FFPE Total RNA Miniprep System provides a complete, all-inclusive method for purification of quality total RNA from formalin-fixed paraffin-embedded tissue without using hazardous solvents or overnight digestion. Total RNA can be isolated from FFPE tissue in approximately one and one-half hours with minimal hands-on time.
Formalin fixation and paraffin embedding (FFPE) is a common method for archiving tissue specimens. The ability to extract RNA from these samples provides the potential for correlating disease state and tissue morphology with gene expression. Historically, extraction of RNA from FFPE tissues has been a challenge because the formalin fixation process cross-links proteins and nucleic acids and introduces modifications to RNA by addition of monomethyl groups to nucleotides. These modifications can inhibit subsequent analysis of RNA isolated from FFPE tissues.
This system also uses a deparaffinization method that does not rely on xylene or other organic solvents. By eliminating the washing steps involved in xylene deparaffinization, this method can help retain small RNA fragments.
Delivery information: Both kit contains enough mineral oil, lysis buffer (LBA), proteinase K solution (PK), MnCl2 (0.09 M), BL buffer, wash solution, DNase I (lyophilized), DNase buffer, nuclease-free water, collection tubes, ReliaPrep FFPE binding columns, and elution tubes to perform total rRNA isolations.
