Dyes for membrane potential measurements, vesicle fusion assays, and for binding to cell membranes or liposomes.

  • Study membrane fusion or permeability
  • Dyes for functional imaging of neurons
  • Membrane potential detection kits

The fluorescence of NBD is highly dependent on the environment; it is virtually nonfluorescent in water but becomes strongly green fluorescent in hydrophobic media

NBD-ethylenediamine can be readily coupled to activated carboxy groups or sulfonyl chlorides. ANTS can be used together with the fluorescent quencher DPX for the studies of membrane fusion or permeability. The mixture of ANTS-DPX is minimally fluorescent initially, but becomes increasingly more fluorescent upon dilution (i.e., membrane fusion or leakage).

APTS is a very useful green fluorescent dye for labeling glycoproteins or sugar molecules in general.

DPH is almost non-fluorescent in water, but it exhibits a strong increase in fluorescence after intercalation into membranes and can be used as a probe for viscosity, polarity and lipid order.

NBD C6-ceramide has been used for following sphingolipid metabolism in cells, and as a selective stain for the Golgi apparatus in live and fixed cells. NBD C6-Sphingomyelin has been used for following sphingolipid metabolism in cells.
Biotin-AM3-25 is a conjugate of AM3-25 and biotin. Membranes labeled with the probe can subsequently couple to avidin or streptavidin.
DPA is a nonfluorescent anionic membrane dye. With its absorbance in the blue region of the visible spectrum, DPA can be used as a FRET quencher for a variety of fluorescent donor dyes.
ANEPPS dyes are fast responding membrane potential dyes. Di-4-ANEPPS has the problem of being internalized in cells over long-term, so it is mainly used for short-term experiments. Di-8-ANEPPS is more lipophilic, and the dye is better retained in the outer leaflet of cell plasma membranes and thus more suitable for long-term membrane potential studies. Di-8-ANEPPS is also more photostable and less phototoxic than Di-4-ANEPPS.DiBAC4(3) is a so-called translational membrane potential dye that redistributes within the cell membrane when membrane potential changes.
Di-2-ANEPEQ , Di-8-ANEPPQ and Di-12-ANEPPQ are fast-responding membrane potential dye. Di-2-ANEPEQ is highly water-soluble and is usually introduced into cells by microinjection. Di-8-ANEPPQ and Di-12-ANEPPQ are more hydrophobic than Di-2-ANEPEQ and useful for potential-sensitive retrograde labeling of neurons.
The styryl dye RH414, RH421, RH237 and RH795 are fast-responding potentiometric probes that are primarily used for functional imaging of neurons.
DiOC5(3) and DiOC6(3) are two of the most widely used carbocyanine dyes used for membrane potential measurements. DiOC2(3) has been used for measuring membrane potentials in bacteria. DiOC6(3) has been used to stain the ER in both live and fixed cells.
In the DPA/Tb3+ vesicle fusion assay, separate vesicle populations are loaded with TbCl3 or DPA. Fusion of the two types of vesicles results in fluorescence increase at 490 nm or 545 nm, with excitation at 276 nm. The Tb3+-DPA complex
10,000 times more fluorescent than free Tb3+. SDIP/Eu3+ as an alternative to DPA/Tb3+. The advantage is that SDIP/Eu3+ generates fluorescent emission with brighter intensity and much longer wavelength. DiO/DPA membrane potential detection kit is comprised of the green fluorescent membrane dye DiOC16(3) and the anionic quencher dye DPA.
The DiO/DPA system detects cytoplasmic membrane potential changes using FRET. DiO is a “stationary” donor while DPA acts as a mobile acceptor.
