McCoy’s 5A Medium is a general purpose medium that supports the propagation of many types of primary cells, established cell lines, and explants from biopsy tissues. McCoy’s 5A Medium uses a sodium bicarbonate buffer system (2.2 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

McCoy’s 5A Medium originated from Dr. McCoy and his coworkers, as it was centered on the amino acid requirements of Novikoff hepatoma cells in culture. This work involved a medium known as Basal Medium 5A and used to grow normal bone marrow, skin, gingiva, testes, mouse kidney, omentum, adrenal glands, lung, spleen, rat embryos and other tissues. This product also includes Dr. Hsu’s addition of Hanks’ salts to enable use outside a CO2 incubator.
McCoy’s 5A Medium with L-glutamine will support growth of primary mammalian cells derived from the following: normal bone marrow skin, spleen, kidney, lung, rat embryos, and other tissues.
