Biotinylated ABL Wild Type Kinase, alias BTN Abl1, Carna Biosciences
Full-length human ABL [2-1130(end) amino acids of accession number NP_005148.2] was expressed as N-terminal DYKDDDK tagged, biotinylated protein (126kDa) using a baculovirus expression system.

FLAG tagged Kinase is purified using DYKDDDDK tag antibody agarose. Amino acid sequence is confirmed by peptide mass fingerprinting. Purity is assessed by SDS-PAGE/CBB staining. Bioactivity is measured using a mobility shift assay. The enzyme is incubated with fluorescently-labeled peptide substrate, Mg++ or Mn++ and ATP. Following the reaction, phosphorylated peptide substrate is separated from unmodified substrate, and detection is performed using a LabChip™3000. Kinase activity is expressed as the percentage of phosphorylated substrate relative to total substrate (% conversion) at a given enzyme concentration within a given time.

The kinase is supplied in a storage buffer of 50mM Tris-HCl, 150mM NaCl, 0.1% CHAPS or 0.05% Brij35, 1mM DTT, 10% glycerol, pH 7.5.
