Dot: Use at a concentration of 1 - 3 ?g/ml - determined using recombinant NGFB at 400 ng under non- and 200 ng under reducing conditions. ELISA: Use at a concentration of 1 - 2 ?g/ml (100 ?l/well) for capture ELISA, Monoclonal Anti-Human Nerve Growth Factor-Beta can be used as the capture antibody. Determined in combination with a biotinylated NGF-Beta polyclonal detection antibody at 50-100 ng/ml (100 ?l/well), an ELISA for sample volumes of 100 ?l with a range of 31.2 to 2000 pg/ml may be obtained. For capture ELISA, a working concentration of 1-2 ?g/ml is determined to coat plates at 100 ?l/well. Inhib: Use at a concentration of 1 - 2 ?g/ml. Human NGFB stimulates the 3H-thymidine incorporation by TF-1 cells in a dose-dependent manner. The Neutralization Dose50 (ND50) for anti-human NGFB is approximately 1-3 ?g/ml in the presence of 5 ng/ml of recombinant human NGFB using the NGFB responsive cell line TF-1. The ND50 is the concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when the cytokine is present at a concentration just high enough to elicit a maximum response. The exact concentration of antibody required to neutralize human NGFB activity is dependent on the cytokine concentration, cell type, growth conditions, and the type of activity studied. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
