ELISA: Use at a concentration of 0.5 - 1 ?g/ml, this concentration detects 0.6 ng/well of rhIL2. IHC: Use at a concentration of 0.5 - 5 ?g/ml to detect IL2 in cultured cells or tissue sections. Neut: Use at a concentration of 0.05 - 0.15 ?g/ml. WB: Use at a concentration of 0.1 - 0.2 ?g/ml, to detect rhIL2 at 5 ng/lane under non-reducing and reducing conditions. Predicted molecular weight: 19 kDa. Anti-Human IL2 is tested for its ability to neutralize the bioactivity of rhIL2 in a cell proliferation assay using an IL2-dependaent murine cell line, CTLL2. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of bioactivity of rhIL2 that is present at concentration just high enough to elicit a maximum response. In this bioassay, rhIL2 is preincubated with various dilutions of the antibody for 1 hour at at 37°C in a 96-well microtiter plate. CTLL2 cells are added to each well. The total volume of 100 ul, containing antibody, rhIL2 at 2 ng/ml and cells at 1 x 10^5 cells/ml, was incubated for 24 hours at 37 °C in a 5% CO2 humidified incubator and then pulsed for the last 4 hours with ^3H-thymidine. Cells were harvested onto glass filters and the ^3Hthymidine incorporation into DNA was measured. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
